| Sanofi-Synthelabo submitted a New Drug
Application (NDA) for oxaliplatin in combination with
5-fluorouracil/leucovorin (5-FU/LV) for the first line
treatment of advanced colorectal cancer in July 1999 to the
FDA, which was based on European studies designed for
acceptable endpoints for approval in Europe. FDA determined
that the NDA met the requirements to be accepted for review
and upon completion of their review held an Oncology Drug
Advisory Committee Meeting to discuss the dossier. At the
advisory committee meeting, it was determined that the dossier
did not meet the U.S. requirements for approval for a
first-line indication, which requires two adequate and
well-controlled studies with a primary endpoint of overall
survival. Sanofi-Synthelabo's NDA consisted primarily of two
Phase III studies, of which neither had overall survival as a
primary pre-specified endpoint. The committee and FDA
acknowledged that oxaliplatin had activity in colorectal
cancer, but based on data from other products to date were not
willing to change the U.S. requirements to an alternative
endpoint to support approval in this stage of the disease.
Sanofi-Synthelabo remains committed to the continued
development and registration of oxaliplatin in the United
States for the treatment of colorectal cancer and other
tumors. After several meetings with the FDA and NCI
representatives, the registration studies for second- and
third-line treatment of colorectal cancer have been initiated.
These stages of disease were selected because they represent
an unmet medical need. Thereby, minimizing the number of
patients needed to evaluate an earlier endpoint of response,
and based on FDA regulations, these unmet medical need stages
will allow accelerated review and approval. |
